Description

Description: This assay employs a two-site sandwich ELISA to quantitate ZC3HC1 in samples. An antibody specific for ZC3HC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ZC3HC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZC3HC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZC3HC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: NIPA, Contains 1 C3HC-type zinc finger.Widely expressed. Highly expressed in heart, skeletal muscle and testis. Expressed in brain, placenta, lung, kidney, liver, pancreas, spleen, thymus, prostate, ovary small intestine and colon. Weakly or not expressed in leukocytes.Essential component of an SCF-type E3 ligase complex, SCF(NIPA), a complex that controls mitotic entry by mediating ubiquitination and subsequent degradation of cyclin B1 (CCNB1). Its cell-cycle-dependent phosphorylation regulates the assembly of the SCF(NIPA) complex, restricting CCNB1 ubiquitination activity to interphase. Its inactivation results in nuclear accumulation of CCNB1 in interphase and premature mitotic entry. May have an antiapoptotic role in NPM-ALK-mediated signaling events
